Flow Cytometric Characterization of Cell Surface Markers to Differentiate Between Fibroblasts and Mesenchymal Stem Cells of Different Origin

Introduction Identification and purification of mesenchymal stem cells (MSCs) expanded in culture for therapeutic use is crucial improved yield optimal results. Fibroblasts are the most common cell type connective tissue commonly found as contaminants MSC cultures, affecting potentially causing tumor formation after transplantation. In current study, we wished to identify surface markers that can differentiate MSCs different origins from fibroblasts. Material methods were isolated bone marrow, adipose tissue, Wharton’s jelly placental fibroblasts foreskin (as a negative control) order examine differences expression panel 14 using multiplex flow cytometry. Results Our results indicate following could be useful differentiating between derived from: - CD79a, CD105, CD106, CD146, CD271; CD14, CD56 CD105; marrow CD146; CD146. Furthermore, that, contradictory previous studies, CD26 not fibroblast-specific. Conclusions conclusion, our study may prove tool discrimination thus used authenticate identity cells.
